چکیده

The effect of isolated cellulose binding domains (CBDs) on the hydrolysis of filter paper and microcrystalline cellulose by both cellobiohydrolase I and endoglucanase, was studied. CBDs were obtained by proteolysis from cellulases using a scaled-up variant of our previous method. Experiments were performed for different enzyme/substrate ratios in both the absence and presence of CBDs. Hydrolysis of filter paper by intact cellobiohydrolase I in the presence of additional CBDs was found to have a synergistic effect, leading to an increase of the sugar production of up to 30%. The effect was less pronounced using microcrystalline cellulose, where an increase up to 16% was observed. Similar trends were found during the hydrolysis of both substrates by endoglucanase. © 2002 Elsevier Science Inc. All rights reserved.

Cellulase hyperproducing mutants derived from the fungus Trichoderma reesei QM9414 produced large amounts of cellulase at the enzymatic and transcriptional levels.

Cellulase hyperproducing mutants derived from the fungus Trichoderma reesei QM9414 were analyzed. They exhibited higher filter-paper degrading activity and a lower growth rate than the wild-type QM9414 strain. Transcription of the cellobiohydrolase I and endoglucanase I genes in the mutants was also greater than that of QM9414, suggesting that cellulase hyperproduction by these mutants was regu...

Catalytic and substrate-binding domains of endoglucanase 2 from Bacteroides succinogenes.

Endoglucanase 2 (EG2) of the cellulolytic ruminal anaerobe Bacteroides succinogenes is a 118-kilodalton (kDa) enzyme which binds to cellulose and produces cellotetraose as the end product of hydrolysis. The purified enzyme was treated with the protease trypsin in an attempt to isolate peptides which retained the ability to either hydrolyze soluble carboxymethyl cellulose or bind to insoluble ce...

Synergistic saccharification, and direct fermentation to ethanol, of amorphous cellulose by use of an engineered yeast strain codisplaying three types of cellulolytic enzyme.

A whole-cell biocatalyst with the ability to induce synergistic and sequential cellulose-degradation reaction was constructed through codisplay of three types of cellulolytic enzyme on the cell surface of the yeast Saccharomyces cerevisiae. When a cell surface display system based on alpha-agglutinin was used, Trichoderma reesei endoglucanase II and cellobiohydrolase II and Aspergillus aculeatu...

Mechanism by which cellulose triggers cellobiohydrolase I gene expression in Trichoderma reesei.

The expression of cellobiohydrolase I mRNA from Trichoderma reesei, measured by Northern blot hybridization, is controlled by the nature of carbon sources used in the culture medium. Cellulose and the soluble disaccharide sophorose, but not glycerol or glucose, act as inducers. Cellobiohydrolase I mRNA was undetectable when antibodies to the major members of the cellulolytic system were present...

Structural and functional studies of fungal cellulose-binding domains by NMR spectroscopy

Cellulose has a important environmental role in the preservation of the global carbon cycle and commercial significance as a raw material for industry. To understand the biodegradation of cellulose on the atomic level, it is important to be able to relate enzyme activities with the three-dimensional (3D) structures of cellulases and cellulose. The cellulolytic system of the fungus Trichoderma r...
